Vitamin B Complex Treatment Attenuates Local Inflammation after Peripheral Nerve Injury

Peripheral nerve injury (PNI) triggers different cellular and molecular events crucial for axon regeneration and tissue reinnervation and inflammation is one of these processes. Macrophage activation is central to PNI pathogenesis, transitioning from a proinflammatory (M1) to an anti-inflammatory (M2) phenotype with distinct functions in inflammation and repair.

This study aimed to assess the impact of vitamin B complex (including B1, B2, B3, B5, B6, and B12) on neuroinflammation, partly regulated by l-type Caᵥ1.2 calcium channels. This study was performed by using a controlled transection of the motor branch of the femoral peripheral nerve in an experimental model. These animals received vitamin B complex injections over various intervals before sacrifice. Isolated nerves were subjected to immunofluorescence analysis.

The results showed that vitamin B complex treatment reduced proinflammatory cytokine expression while increasing anti-inflammatory cytokine expression, and this led to neuroinflammation resolution. B vitamins decreased the number of M1 macrophages expressing the Caᵥ1.2 channel and increased the number of M2 macrophages expressing the same channel, suggesting a role in M1/M2 transition post-PNI.

The conclusion is that B vitamins can be used in treating neuroinflammation and promoting neuroregeneration, potentially serving as an effective therapy for PNI in humans.
